Resistance to colistin in Acinetobacter baumannii associated with mutations in the PmrAB two-component system.

چکیده
The mechanism of colistin resistance (Col(r)) in Acinetobacter baumannii was studied by selecting in vitro Col(r) derivatives of the multidrug-resistant A. baumannii isolate AB0057 and the drug-susceptible strain ATCC 17978, using escalating concentrations of colistin in liquid culture. DNA sequencing identified mutations in genes encoding the two-component system proteins PmrA and/or PmrB in each strain and in a Col(r) clinical isolate. A colistin-susceptible revertant of one Col(r) mutant strain, obtained following serial passage in the absence of colistin selection, carried a partial deletion of pmrB. Growth of AB0057 and ATCC 17978 at pH 5.5 increased the colistin MIC and conferred protection from killing by colistin in a 1-hour survival assay. Growth in ferric chloride [Fe(III)] conferred a small protective effect. Expression of pmrA was increased in Col(r) mutants, but not at a low pH, suggesting that additional regulatory factors remain to be discovered.

Stereochemical Trajectories of a Two-Component Regulatory System PmrA/B in a Colistin-Resistant Acinetobacter baumannii Clinical Isolate
Background: There is limited information on the 3D prediction and modeling of the colistin resistance-associated proteins PmrA/B TCS in Acinetobacter baumannii. We aimed to evaluate the stereochemical structure and domain characterization of PmrA/B in an A. baumannii isolate resistant to high-level colistin, using bioinformatics tools. Methods: The species of the isolate and its susceptibility ...
متن کاملGlobal Epidemiology on Colistin Resistant Acinetobacter baumannii
While colistin remains a last resort agent to treat multi-drug resistant Acinetobacter baumannii, resistance to colistin has been reported throughout the world. The resistance has been attributed to mutations in lipid A biosynthesis genes and point mutation in PmrAB-two component response regulator and sensor kinase system. The emergence of plasmid mediated colistin resistance (mcr-1), in multi...
متن کاملThe Effect of Colistin Resistance-Associated Mutations on the Fitness of Acinetobacter baumannii
Acinetobacter baumannii had emerged as an important nosocomial and opportunistic pathogen worldwide. To assess the evolution of colistin resistance in A. baumannii and its effect on bacterial fitness, we exposed five independent colonies of A. baumannii ATCC 17978 to increasing concentrations of colistin in agar (4/5) and liquid media (1/5). Stable resistant isolates were analyzed using whole g...

for gen. sp. 13BJ). The most noteworthy were the results for A. beijerinckii, A. junii and gen. sp. 13BJ. Whereas the former two species included both susceptible and resistant strains all eight strains of gen. sp. 13BJ yielded MICs 16 mg/L. It has recently been shown that therapy with colistin can be compromised by the selection and spread of colistin-resistant A. baumannii strains. Our data f...

Two different mechanisms of resistance to colistin in Acinetobacter baumannii have been described. The first involves the total loss of lipopolysaccharide (LPS) due to mutations in the lpxACD operon, which is involved in the lipid A biosynthesis pathway. The second entails the addition of ethanolamine to the lipid A of the LPS resulting from mutations in the PmrAB two-component system. To evalu...
